Yes, spelling does have a lot to do with it in some cases. For example, I see Jesse as male, Jessie as female. Same with Lee / Leigh, Sidney / Sydney, Ray / Rae, and Loren / Lauren. However, with some names it doesn't matter at all. Spelling Murphy or Harley with an -ie instead of a -y won't make them any more feminine in my mind.
